Polyethyleneimine (PEI) is a synthetic, water soluble, linear or branched polymer. It is widely used in gene delivery system due to the ability to build a complex with DNA and support the release of endosomal through “proton sponge effect.” It is also facilitating the intracellular transport into nucleus.

Is PEI negative or positive?

High positive charge on the surface of PEI leads to electrostatic interaction with the negatively charged components of the cell membrane and causes membrane disruption and damage (6).

Is PEI positively charged?

Polyethylenimines. Polyethylenimines (PEIs) are the second most frequently used polymer for the preparation of DNA polyplexes. Their strong positive charge given by the high nitrogen content in the backbone of the molecule leads to undesirable side effects.

What does PEI do in transfection?

As with most transfection reagents, PEI-mediated transfection involves the formation of nanoparticles (polyplexes) which are obtained by its mixing with plasmid DNA. A short incubation period that allows polyplexes to reach their optimal size is performed prior to their addition to the culture.

How long is PEI transfection?

Transfection with PEI requires two incubation periods; one is when you mix the PEI with your medium (let it rest for 5 min) and the other one is when you mix your DNA with you PEIed medium (20 minutes at least).

How do you transfect cells with PEI?

Gently add the diluted PEI to the diluted DNA. Add the diluted PEI dropwise while gently flicking the diluted DNA tube. Incubate the mixture 15-20 min at room temperature. Carefully transfer the transfection mix to the Lenti-X 293T packaging cells.

How long is PEI good for?

two months
It should be good for two months at least. But watch the transfection efficiency closely after two months. If you think there is a decline in transfection efficiency, discard and get a new tube.
